The German microbiologist, who discovered the diphtheria-causing organism, Corynebacterium diphtheriae, was first observed by German physiologist Theodor Klebs in Diphtheria patient’s throat. Isolation and culturing purely cultures involves some serious technical problems; Löffler realized there was a need to develop a new thick, medium serum, as the conventional gelatin used by Robert Koch required too low a temperature for the diphtheria pathogen.
